microRNA Ready-to-Use PCR panels

Pre-aliquoted microRNA PCR primer sets in 384-well PCR plates for exceptionally sensitive and specific expression profiling. Profile up to 752 human microRNAs using only 40ng total RNA – no pre-amplification necessary. Panels are available for human, mouse and rat microRNAs.

Features

The microRNA Ready-to-Use PCR panels enable exceptionally sensitive high-throughput expression profiling from minimal amounts of starting material. The combination of a Universal cDNA synthesis reaction and Ready-to-Use PCR panels enables fast and easy microRNA profiling. The panels contain dried down primers for one reaction per well and simply require addition of cDNA and ExiLENTSYBR® Green master mix before real-time PCR amplification.

The LNA™-enhanced, microRNA specific PCR primers offers 1) exceptional sensitivity as well as extremely low background enabling accurate quantitation of very low microRNA levels and 2) highly specific assays that allow discrimination between closely related microRNA sequences. Coverage

Two panels covering 752 human microRNAs and two panels covering 752 mouse and rat microRNAs are available. Panel I for human and mouse/rat contains high-priority microRNA primer sets. These microRNAs are generally more highly expressed, more likely to be differentially expressed in disease or more often cited in the literature. Panel II contains primer sets for the most important microRNAs not available on Panel I. To view the list of primer sets in each panel, please download the plate layout files .

The miRNome panels include 6 potential reference gene primer sets , three small RNA reference genes (U6, SNORD38B and SNORD49A for human and U6 snRNA, RNU5G, RNU1A for mouse and rat) and three microRNA reference genes (miR-103-3p, miR-191-5p and hsa-miR-423-5p or mmu-miR-423-3p) all found on Panel I. In addition, Panel I also contains qPCR assays for the 5 synthetic RNAs in the RNA Spike-in kit (cel-miR-39-3p, UniSp2, UniSp4, UniSp5 and UniSp6) that may be used for monitoring the cDNA synthesis or RNA purification (UniSp6 is also provided in the cDNA Synthesis kit II). Finally, each panel contains an inter-plate calibrator in triplicate and an empty negative control.


V4 Update

The updated panels include 54 updated assays un Panel I and 33 updated assays on Panel II. The assays target the same microRNAs as v3 panels. Furthermore the two spike-in primer sets UniSp5 and UniSp6 have been updated for improved performance. Their respective target templates will be the same.

The microRNA Ready-to-use PCR panels have been optimized for use with the Universal cDNA synthesis kit II and ExiLENT SYBR® Green master mix kit. Use of other reagents will affect the quality of the results.

Accurate microRNA expression profiling with minimal sample

The microRNA Ready-to-Use PCR panels enable profiling of 752 microRNAs or both Human panel I and II, from just 40ng total RNA. This makes it possible to do high quality microRNA expression profiling in samples that contain very little total RNA, such as FFPE sections and serum/plasma (Figures 1 and 2).

The only platform with perfect specificity

The miRCURY LNA™ Universal RT microRNA PCR System is the only microRNA platform with absolute specificity (see the miRQC study published in Nature Methods) and out-performed a competitor probe-based microRNA qPCR platform in specificity tests (Figure 3). Perfect specificity eliminates false positives and ensures only robust, reliable microRNA signals.

Fast, easy and reproducible

Save time and effort in the laboratory with the 3 hour easy-to-follow protocol. The synthesized cDNA is simply diluted, combined with the ExiLENT SYBR® Green master mix and aliquoted into all the wells of the 384-well PCR plate which is then ready to run in the real-time PCR instrument. The number of pipetting steps is reduced to a minimum and technical variation is minimized. As a result, it is possible to achieve extremely high reproducibility from day-to-day (Figure 4) and even site-to-site (Figure 5). Very high levels of reproducibility are even possible when profiling using difficult samples such as RNA purified from serum (Figure 6).

A unique system developed specifically for microRNA profiling

The miRCURY LNA™ Universal RT microRNA PCR System offers the best available combination of performance and ease-of-use on the market because it unites two important features (Figure 2):
  • One cDNA reaction for all microRNAs – One single Universal first strand cDNA synthesis reaction is used as template, regardless of the number of microRNAs being profiled. This saves precious sample, reduces technical variation, means less pipetting and saves time in the laboratory.
  • Two LNA™-enhanced microRNA qPCR primers – Both qPCR primers are microRNA-specific and optimized with LNA™. LNA™ primers bind with high affinity, and are shorter than standard PCR primers, so that both primers can fit on the microRNA without overlapping. The result is unrivalled sensitivity and specificity, and extremely low background.

Unmatched sensitivity

Universal RT combined with LNA™-enhanced and Tm normalized primers enables accurate and reliable quantification of individual microRNAs from as little as 1 pg total RNA (Figure 3). In comparison to other microRNA real-time PCR systems using either stem-loop or standard DNA primers, the LNA™-enhanced primers offer significantly increased sensitivity, especially for AU-rich microRNAs.

Exceptional sensitivity as well as extremely low background enables accurate quantification of very low levels of microRNA without the need for pre-amplification. This makes the miRCURY LNA™ Universal RT microRNA PCR System suitable for all sample types, and especially samples with low RNA content e.g. biofluids such as serum/plasma.

Fully validated and optimized

All miRCURY LNA™ Universal RT microRNA PCR primer sets have been optimized for maximum sensitivity and thoroughly validated either by wet lab testing or in silico. In wet lab validation, over 95% of assays detect 10 microRNA copies or less in the PCR reaction (Figure 4). The primer sets have also been validated for specific amplification of the target and for minimal background signal.

Truly specific – no false positives

The incorporation of LNA™ in the qPCR primers facilitates the design of assays that can distinguish between microRNA sequences that differ by a single nucleotide. This makes it a truly specific microRNA qPCR platform – the only microRNA platform that gives no false positive signals (Figure 5). In addition, the assays can discriminate between mature microRNA sequences and precursor microRNA (Table 1).

Fast, easy and reproducible

Save time and effort in the laboratory with the 3 hour easy-to-follow protocol that minimizes pipetting. By using the same cDNA as template in all subsequent PCR reactions, the procedure is greatly simplified compared to systems that require microRNA-specific cDNA synthesis.

With fewer pipetting steps, technical variation is also reduced. As a result, it is possible to achieve extremely high reproducibility from day-to-day (Figure 6) and even site-to-site.

Flexible qPCR system

Tailor your experimental setup to your specific need using individual assays or fully flexible custom Pick-&-Mix panels . Using the same cDNA reaction, shift seamlessly between individual primers, pre-designed miRNome and Focus panels, or custom Pick-&-Mix panels.
