Features

The miRCURY™ RNA Isolation Kit- Biofluids is a high quality kit optimized for the purification of small RNAs from serum, plasma and other biofluids. Based on spin column chromatography, the kit uses a proprietary resin as the separation matrix without the use of toxic phenol or chloroform.

Use the kit to isolate all RNAs smaller than 1000 nt, from mRNA and tRNA down to microRNA and small interfering RNA (siRNA), from other cell components, such as proteins.

Take advantage of the optimized yet fast and easy-to-use protocols based on either centrifugation or a vacuum manifold. The workflow is outlined in Figure 1.

RNA isolated using the kit is fully compatible with the miRCURY LNA™ Universal RT microRNA PCR System and ideal for highly sensitive quantification of microRNA in applications such as biomarker discovery and toxicology studies.


Coverage

The miRCURY™ RNA Isolation Kit- Biofluids is optimized for handling low-yield samples containing high concentrations of inhibitors and should therefore be well suited for biofluids in general. Optimized protocols and recommendations are available for serum/plasma, urine and CSF.

Although not species specific, the protocols are optimized for working with human biofluids. Recommended starting material for different organisms and sample types can be found in the manual.

The Biofluids kit is available in two versions that contain spin columns and regents for either 10 or 50 small RNA purifications.

Superior yield and quality

Purification of microRNA from biofluids is challenging due to the often very limited RNA content and high levels of qPCR inhibitors. The miRCURY™ RNA Isolation Kit- Biofluids is specifically optimized for handling low yield samples and for reducing inhibitor carry-over to a minimum.

The superior RNA yield and purity possible with the miRCURY™ RNA Isolation Kit – Biofluids is demonstrated in Figures 1 and 2 where qPCR –based detection of microRNAs from plasma has been performed using four different isolation kits.


High purity and reproducibility

Robust and reproducible purification of RNA is particularly important from challenging samples such as biofluids that are subject to many pre-analytical variables.

The miRCURY™ RNA Isolation Kit - Biofluids uses a simple column-based procedure without the need for toxic phenol or chloroform. This leads to high purity RNA and high reproducibility between individual isolations (Figure 3). In addition, we recommend the use of carrier RNA to increase reproducibility for samples with low microRNA content (see Figure 4).

In comparisons with three other commercially available RNA isolation kits, the miRCURY™ RNA Isolation Kit – Biofluids gives the lowest standard deviation in Cq values between different RNA purifications from the same plasma sample (Figure 5).


Flexibility

The miRCURY™ RNA Isolation Kit – Biofluids can be used for RNA purification from a variety of different biofluids. Optimized protocols and recommendations are currently available for serum/plasma, urine and CSF. Recommended starting material for different organisms/sample types can be found in Table 1.
