Exiqon's miRCURY™ LNA Knockdown technology enables sequence-specific inhibition of mature miRNAs in vitro and in vivo. Depletion of miRNAs can be judged by the decrease in hybridization signals in Northern blots as LNA-miRNA hybrids are thermally stable in denaturing gels.
Exiqon uses in-house bioinformatics expertise to optimally design miRCURY™ LNA Knockdown probes for specific knockdown of miRNAs.
miRCURY™ LNA Knockdown probes have high potency, without compromising specificity. Effective knockdown of miRNA is achieved with standard or low concentrations of miRCURY™ LNA Knockdown probe. However, this knockdown is reduced significantly with the presence of a mismatch between the probe and the target. Our data demonstrates that the presence of two mismatches between a miRCURY™ LNA Knockdown probe and its target is sufficient to completely eliminate inhibition. This is important within the cell, where closely related miRs may be present and the knockdown needs to be as specific as possible.
miRCURY™ LNA Knockdown probes have also been demonstrated to be more effective than standard methods used in miRNA knockdown, such as DNA-based probes and those based on 2’-O-methyl chemistry.
The inclusion of LNA™ in Exiqon’s miRCURY™ LNA Knockdown probes provides improved resistance to cellular nucleases, thus providing more lengthy periods over which to conduct your knockdown studies.
