ConsultationWhen you place an order for Exiqon's OptiScreen Service, you are guaranteed direct communication with the scientists performing your experiments throughout the duration of the project. You will be asked to submit a sample submission form. Following this step, you will receive a complimentary consultation with experts who have a wealth of experience in microRNA in situ Hybridization. At this consultation, the optimal experimental design and is discussed and agreed upon.
Tissue Samples Exiqon’s OptiScreen Service is currently available for human or mouse FFPE tissues. Customers are asked to submit FFPE blocks, which will be sectioned by Exiqon, according to strict guidelines and procedures.
For good results, it is very important to use high quality tissue samples. At the initial consultation we will offer advice on methods for harvesting and preservation of de novo samples. If possible, it is therefore ideal if the consultation takes place before you obtain your tissue samples. Please find more details on sample preparation under the “Sample submission” section of the workflow above. |

Sample SubmissionAfter discussing the experimental design with our scientists, you will be asked to complete the sample submission form. Technical specialists will review the information to make sure that there are no outstanding questions and will then ask you to ship your samples. Whether you wish to submit only a few or several hundreds of samples we are happy to accept your order. |

Quality controlAfter receiving your tissue samples, our specialists will assess the integrity of select blocks in a full ISH setting, using positive and negative controls. In addition, all detection probes will be subjected to a strict quality control step, prior to application to your samples. Any issues related to sample quality will be immediately brought to your attention, with accompanying data, to illustrate the situation, and help determine the best path forward. |

HybridizationAll Exiqon OptiScreen projects feature our pre-designed, LNA™-enhanced Double-DIG* detection probes, for assured performance and maximum sensitivity. All detection probes are subjected to a strict quality control process prior to application to your submitted samples.
A multi-variable optimization step will be performed on submitted blocks, to determine the best conditions for detecting the microRNA species requested. Sections will be tested under a variety of probe concentrations, hybridization temperatures, and proteinase conditions. This optimization is followed by high-throughput imaging of all microRNAs of interest, under the best determined conditions, along with positive and negative controls. You will receive, along with your results report, all details regarding the various conditions tested, including images obtained during both optimization and screening phases.

DeliverablesWhen the service order has been completed you will receive an e-mail notifying you to expect delivery of your results package. The Exiqon OptiScreen Results Report will contain a description of the project and an overview of the results and an assessment of the quality of the data.
You will be able to download all obtained images from our server at 20X magnification. In addition to the Results Report and images, you will also receive all unused block portions, as well as slides utilized in the service run. Our technical experts will remain at your disposal for guidance and assistance in the interpretation of results as needed, ensuring the highest level of attention to your specific project needs. |
