Cancer Focus microRNA PCR Panels

PCR panels containing miRCURY LNA™ Universal RT microRNA PCR assays for microRNAs related to cancer. Panels are available in ready-to-use 96 well plates – just add cDNA and PCR master mix. New v4 panels with optimized assays for improved specificity.

microRNA and cancer

There have been numerous studies demonstrating links between microRNA dysregulation and various disease states, including a diverse array of human cancers. MicroRNAs have been shown to act as both oncogenes and tumor suppressors.

The high relative stability of microRNA in common clinical source materials (e.g. FFPE blocks, plasma, urine, saliva, etc.) and the ability of microRNAs to function as molecular markers for discrete tissue types and disease states have positioned microRNA quantification as a promising new tool for a wide range of diagnostic applications. MicroRNAs can be used as disease classifiers for different types of cancers or as biomarkers to monitor the effects of therapy on cancer prognosis and progression.

A unique system for microRNA profiling

The miRCURY LNA™ Universal RT microRNA PCR offers the best available combination of performance and ease-of-use on the microRNA real-time PCR market because it unites two important features (Figure 1):
  • Universal RT – One first-strand cDNA synthesis reaction (or RT reaction) can be used as template for multiple microRNA real-time PCR assays. This saves precious sample, reduces technical variation and saves time in the laboratory.
  • LNA™ PCR amplification – Both PCR amplification primers (forward and reverse) are microRNA specific and optimized with LNA™. The result is 1) exceptional sensitivity as well as extremely low background enabling accurate quantitation of very low microRNA levels and 2) highly specific assays that allow discrimination between closely related microRNA sequences.

Unmatched sensitivity

The exceptional sensitivity of the microRNA PCR primer sets is achieved by combining the following features:
  • Universal RT
  • LNA™ enhanced and Tm normalized primers
This combination enables accurate and reliable quantification of individual microRNAs from as little as 1 pg of total RNA input in the initial first-strand cDNA synthesis (Figure 2).
In comparison to other microRNA real-time PCR systems using either stem-loop or standard DNA primers, the LNA™-enhanced primers offer significantly increased sensitivity, especially for AU-rich microRNAs.


Fully validated and optimized

All miRCURY LNA™ Universal RT microRNA PCR primer sets have been optimized to be as sensitive as possible. Over 80% of assays detect minimum 100 microRNA copies in the PCR reaction whereas close to 50% detect as little as 10 microRNA copies (Figure 3). The primer sets have also been validated for specific amplification of the target and for minimal background signal.

Excellent specificity

The incorporation of LNA™ in the PCR amplification primers (forward and reverse) facilitates the design of assays that can distinguish between microRNA sequences that differ by a single nucleotide. In addition, the assays can discriminate between mature microRNA sequences and precursor microRNA (Table 1).


Fast, easy and reproducible

Save time and effort in the laboratory with the 3 hour easy-to-follow protocol. By using the same RT reaction as template in all subsequent PCR reactions, the procedure is greatly simplified compared to systems that require microRNA-specific first-strand synthesis. The number of pipetting steps is reduced to a minimum and technical variation is minimized. As a result, it is possible to achieve extremely high reproducibility from day-to-day and even site-to-site.

Use Exiqon reagents

The microRNA Ready-to-use PCR panels have been optimized for use with the Universal cDNA synthesis kit and SYBR Green master mix kit. Use of other reagents will affect the quality of the results.

Cancer Focus microRNA PCR Panel

Exiqon’s Cancer Focus microRNA PCR Panel targets 84 human microRNAs related to cancer. These microRNAs have been shown to be important for tumorigenesis and in cancer diagnostics.

These microRNAs can be used as markers for 27 different types of cancer (Table 1). They have been identified from a large biobank containing thousands of FFPE samples as well as from literature data. For a list of references of each microRNA on the panel, please refer to the plate layout file.

The panel is designed for cancer microRNA analysis of cancer progression, recurrence and metastasis and will help researchers to quickly and accurately analyze cancer samples and help identify new microRNA-based biomarkers.

Product coverage

Cancer Focus microRNA PCR Panels are available in:
  • 96 well plate format: 4 sets of 96-well plates with one Cancer Focus Panel (sufficient for 4 samples).
  • 384 well plate format: Eight Cancer Focus Panels delivered in two 384 well plates (sufficient for 8 samples). These plates have been specifically designed for fast and easy manual and multi-pipetting.
Both formats are Ready-to-use with 10µl reactions per well.

The panel contains assays for the following:
  • 84 microRNAs specifically linked to cancer
  • 3 microRNA reference genes: miR-103-3p, miR-191-5p, miR-423-5p
  • snRNA reference genes: SNORD49A, U6 snRNA, SNORD38B
  • 3 Inter-plate calibrators (UniSp3 IPC)
  • 5 RNA spike-in (cel-miR-39-3p, UniSp2, UnuSp4, UniSp5 and UniSp6)
The Cancer Focus microRNA PCR Panels are compatible with most real-time PCR instruments (Figure 1).

Cancer Focus microRNA PCR Panels can be used for microRNA profiling for many types of cancer from clinical samples including FFPE and biofluids. These microRNA profiles can be used to identify unique cancer signatures and help researchers and clinicians to identify biomarkers or classifiers for cancer types and progression (Figure 1 & 2).
