mRNA in situ hybridization

Custom LNA™ detection probes for in situ hybridization and Northern blotting are available in a "ready to label" format (for use in combination with enzymatic labeling kits) and in a number of pre-labeled formats, e.g. labeled with DIG, biotin etc. SEE TABLE BELOW FOR AVAILABLE LABELS AND PRICES.

The probes are designed using a in-house software which is based on the extensive experience of Exiqon’s scientists in LNA™ probe design. This software has proven very successful in providing probes with:
                   - Optimal LNA™ content and positioning
                   - Optimal Tm
                   - Minimal self-annealing properties
                   - Optimal target recognition (BLAST search is included in the service when relevant)

To request a probe design, please provide Exiqon with the target sequence (mRNA sequence) and your choice of label in this form and we will get back to you within 24 hours. Please also include any comment you might have regarding the design of the probe (e.g. in which region of the sequence you would prefer the probe to target).

A number of predesigned control LNA™ mRNA in situ hybridization probes are available. These probes have similar length and LNA content as the antisense detection probes.

IMPORTANT NOTE: In order to achieve the full benefit of an LNA Detection probe high hybridization stringency is needed. As a general guideline hybridizations should be performed at 21°C below the calculated Tm of the LNA probe in 50% formamide hybridization buffer.
