Coverage The library targets 722 human microRNAs listed in miRBase v. 11.0. In addition, the library targets 36 human proprietary miRPlus™ microRNAs not yet in miRBase.

Designed for genome-wide high throughput screening The miRCURY LNA™ microRNA Knockdown library enables genome-wide high throughput screening of microRNA function. The knockdown probes in this library have been specially designed for this purpose.
With large screening projects involving >700 knockdown probes uniform transfection conditions must be adopted for reasons of practicality i.e. the same amount of each probe in the library is used for all the transfections. For this reason it is crucial that all the knockdown probes have similar potencies towards their intended microRNA target. This is not the case with libraries based on traditional antisense inhibitors (for example 2’-OMe oligonucleotides) since the potency of such inhibitors is dictated to a large extent by the GC content of their target microRNA.
We have solved this problem by carefully designing the length, sequence and LNA™ spiking patterns of our knockdown probes. Uniform high potency of all the microRNA inhibitors in the library was accomplished by ensuring Tm normalization around an optimal temperature while keeping the level of self-complementarity to a minimum.
High potency and purity minimizes side effects Due to the high potency of novel designed knockdown probes they can be used efficiently at low concentrations. This minimizes problems with secondary effects, not related to antisense inhibition of the microRNA target, which may confound the phenotypic readout from the cellular assay. The probes are HPLC purified and desalted after purification. The high purity of the probes ensures minimal side effects from toxic byproducts.
Format The library is delivered in nine 96-well plates. Each well contains 0.25 nmol dried down miRCURY LNA knockdown probe. Plate layout is available as a downloadable Excel file.
