| | Reliable microRNA profiling results can be obtained from only 30 ng of total RNA. The log expression ratios of Tumor vs. Normal adjacent tissue were conserved for most microRNAs across the different total RNA input amount tested (30 ng to 1000 ng). For some microRNAs (e.g. miR-93, -98), the log ratios show no variation at low input compared to higher input amounts. Figure 2a. Heat map of esophageal cancer. Log expression ratio tumor vs. normal (T/N) adjacent tissue are shown using various amounts of total RNA. T/N 30 ng, T/N 100 ng, T/N 300 ng and T/N 1000ng is the log ratio of tumor vs. normal using 30 ng, 100 ng, 300 ng and 1000 ng total RNA, respectively. Data is average from two experiments. Figure 2b. Excellent correlation of 91% between 30 ng and 1000 ng total RNA samples. Log2 ratios of tumour vs. normal (T/N) adjacent tissue of a oesophagus cancer using 1000 ng input material is plotted against Log2 ratios in identical experiments using 300 ng, 100 ng, and 30 ng total RNA.
