microRNA profiling with as little as 200ng total RNA

Our miRCURY LNA™ microRNA Array platform is the most sensitive platform on the market. Due to the incorporation of high affinity LNA™, more than 90% of our probes have a detection limit ≤ 10amol. This enables us to perform microRNA profiles with very low levels of total RNA input.

However, there is no simple answer to the question of how much RNA is needed to perform microRNA profiling. It depends very much on the type of experiment and the scientific questions being asked. Below a certain threshold level using less input RNA comes at the expense of lost signals from low level expressed microRNAs. With our platform, we observe a gradual and moderate drop in call rate when hybridizing with less than 1µg total RNA. Therefore, if you wish to detect even the weakest expressed microRNA we recommend using total RNA samples of 1-2 µg depending on the experimental setup (half the sample may be used for a common reference or in a dye swap). However, we can generate excellent and biologically significant results using 1/10 of this amount (or even less when comparing samples that are very different. See Figure 1.). Considering that 100ng is consumed for RNA quality control and quantification this enables us to perform services with samples of only 200-300ng total RNA.

We perform dual color experiments because this approach offers significant advantages in terms of comparison of data from individual arrays.
